Place a prepared slide on the stage and secure it with the stage clips or mechanical stage. Start with the lowest magnification (100×) to locate the area of interest and use coarse focus to bring the specimen into view, then refine with fine focus. Switch to higher magnification (200× or 450×) as needed to examine finer details, making small focus adjustments each time. Adjust the illumination intensity to improve contrast and clarity for translucent samples. When finished, lower the stage, remove the slide, turn off the light source and cover the microscope if storage is required.
For reliable results, use properly prepared and clean slides, avoid excessive light intensity that may wash out details, and always handle optical components by their edges. Regularly clean lenses with appropriate lens tissue and store the instrument in a dry, dust-free place to maintain optical performance.
